Air samples were eluted as described previously (19). Dust was collected from petri dish base and lid by repeated pipetting with 2?ml extraction buffer (PBS, 0.5% Tween 20, 0.15% Kathon CG, 1% BSA). Samples were rotated for 2?h, centrifuged at 1000?g for 10?min, and supernatants were stored at ?20C. Air and petri dish samples were analyzed for rat urinary allergen Rat n 1 (RUA) and mouse urinary allergen Mus m 1 (MUA) by ELISA assay, as described previously (19, 21). Analysis of cat allergen (Fel d 1) was performed using commercial reagents (Indoor Biotechnologies, Charlottesville, VA, USA) as described (20). Samples near or below the limit of detection ( recommendations. Detection [http://www.selleckchem.com/products/sch58261.html find more] limits were 10?pg/ml or about 20?pg/m3 for rodent allergens and 5?pg/ml (10?pg/m3) for cat allergen, respectively. Air filters were extracted in 2?ml sterile water and 0.05% Tween 20 in sterile Falcon tubes (2085; Becton Dickinson, Stockholm, Sweden), rotated for 1?h, and centrifuged at 1000?g for 10?min to remove debris. The supernatants were stored in sterile cryotubes (Nunc A/S, Roskilde, Denmark) at ?20C. A colorimetric limulus amebocyte lysate [http://en.wikipedia.org/wiki/Thymidine_kinase Thymidine kinase] (LAL) test QCL-1000 (Bio Whittaker, Walkersville, MD, USA) was used to determine endotoxins as described by the manufacturer. The detection limit was 0.33?EU/m3 at 5?h of air sampling. Main outcome variables were work-related allergic symptoms and sensitization (SPT or IgE antibodies) to pet shop allergens. Mean personal airborne and sedimenting dust RUA and MUA levels, respectively, and airborne endotoxin values for each shop were allocated to each study participant from that shop. Analyses were mainly performed using statview (v 5.01; sas Institute, Solna, Sweden). Values that were not normally distributed are presented as median with 25C75th percentiles. Chi-square test (or Fishers exact test) was used to test correlation between factors. Nonparametric tests were used to test exposure differences between groups. To analyze risk factors and presence of work symptoms or sensitization, for dichotomous variables, prevalence ratios (PR) with 95% confidence interval (CI) were calculated (EpiInfo, [http://www.selleckchem.com/products/q-vd-oph.html Selleck Q VD Oph] OpenEpi v 2.3; CDC, Atlanta, GA, USA). For continuous variables, MannCWhitney U-test test was used. P?
